TALEN表达载体构建技术研究进展

Progress in Construction of TALEN Expression Vectors

  • 摘要: 转录激活因子样效应蛋白核酸酶(TALEN) 是最新出现的基因组定点修饰技术之一。该技术在应用过程中为保证靶点序列的特异性, 编码转录激活因子样效应物(TALE)结构域的序列一般较长,这使得TALEN表达载体的构建成为该技术的一个关键步骤和应用中的主要瓶颈,为此发展了多种TALEN表达载体构建方法。本文主要对其中常见的Golden Gate法、REAL法、单元组装法、FLASH法、ICA法、LIC法等方法及其研究进展进行了总结。随着TALEN表达载体构建方法的进一步丰富和普及,该技术必将在生物基因功能研究、农作物和家畜性状改良、人类遗传疾病治疗方面展现出广阔的应用前景。

     

    Abstract: Transcription activator-like effector nuclease (TALEN) was one of the latest genome modification technologies, which was an artificially engineered hybrid protein. To ensure the specificity for the target sequence, sequences encoding transcription activator-like effectors (TALE) domains generally was so long. Therefore, construction of TALEN expression vectors was a key step in the application of TALEN. This paper introduced construction methods of TALEN expression vector, including Golden Gate, REAL, Unit Assembly, FLASH, ICA, LIC and so on, and the latest research progress. With construction methods of TALEN expression vector further enriched and popularized, this technology had broad application prospects in the biological study of gene function, crops and livestock improvement, the treatment of human genetic diseases.
