基于响应面法优化柱前衍生−高效液相色谱法分析传统郫县豆瓣中8种生物胺

Analysis for Eight Biogenic Amines in Traditional Pixian Broadbean by a Pre-column Derivatization-HPLC Method Based on Response Surface Optimization

  • 摘要: 在通过传统工艺发酵的过程中,郫县豆瓣存在生物胺污染的潜在风险。本文以丹磺酰氯(dansyl chloride, Dns-Cl)为衍生剂,通过衍生剂浓度、衍生时间和温度、体系pH值和提取溶剂进行响应面法优化,确定了郫县豆瓣中生物胺(biogenic amines,BAs)柱前衍生−反相高效液相色谱法(RP-HPLC)的最优分析条件,并在该条件下对3个不同发酵年份的郫县豆瓣中的组胺、酪胺、尸胺、腐胺、苯乙胺、色胺、精胺和亚精胺8种BAs进行了实际样品分析。结果表明,衍生时间38 min、衍生温度41 ℃、体系pH=9、Dns-Cl质量浓度11 mg/mL为最优分析条件。在该条件下,方法检测郫县豆瓣中BAs的线性范围为1~500 mg/L,相关系数R2≥0.997 7,检出限0.1~0.6 mg/L,定量限0.4~2.0 mg/L,方法加标回收率为90.44%~109.29%,相对标准偏差(RSD)为1.02%~2.54%。综上所述,该方法灵敏度高、重现性好且结果准确可靠,可用于郫县豆瓣中BAs的检测。

     

    Abstract: A potential risk of biogenic amine contamination exists in Pixian broadbean due to its the traditional fermentation process. The concentration, time and temperature of dansyl Chloride (Dns-Cl) derivation, as well as reaction system pH and extraction solvent were optimized by response surface methodology in order to establish the pre-column derivatization reversed-phase high performance liquid chromatography (RP-HPLC) for biogenic Amines (BAs) analysis. Under these optimum conditions, eight kinds of biogenic amines, including histamine, tyramine, cadaverine, putrescine, phenethylamine, tryptamine, spermine and spermidine in Pixian broadbean samples with three different fermentation times were analyzed. The results show that the optimum conditions are: Dns-Cl concentration is at 11 mg/mL, and derivatization conditions are 38 min at 41 °C, and pH is 9. Under these conditions, each biogenic amine in the Pixian broadbean could be detected in a good linear correlation covering the concentration range of 1 ~ 500 mg/L, (R2 ≥0.997 7). The detection limits ranged from 0.1 to 0.6 mg/L, and the limit of quantification ranged from 0.4 to 2.0 mg/L. The recovery rates were 90.44% to 109.29%, with relative standard deviations (RSD) ranging from 1.02% to 2.54%. In conclusion, the developed method owns high sensitivity, good reproducibility and accurate and reliable results.It can be used for the detection of biogenic amines in Pixian broadbean.
